You can but take in mind that Oni must be a guy. That's something I forgot to mention. Other ancestors change their genders however Ryuu has always remained female, but Oni - male.

I'm also wondering what role a human plays in this. Are they important (well obviously) or like do they have any special connections with these clans other than the fact that the Oni clan doesn't like them?

Well, humans role is to prove to God descendants that they are not all hopeless and well dumb as Oni had thought in the beginning. Originally humans know nothing of these clans power however they do know the legend.

No, since your character is just a descendant so she's still part human. Hebi however did have wings since he was actually a Quetzalcoatl.

I guess since the Oni role would be taken all that would be left would be the human. Though judging from the title of this RP it would be required for the human to be a female and I just don't really RP as them.
