Sekirei: It takes place in 2020 in the fictional city of Oasis . The Sekirei are extraterrestrial super powered beings with a genetic code similar to humans. The Sekirei spaceship crashed on Earth in 1999 and was found by Michael Scarletti (President of Scarletti Enterprises or S.E.) and Marguerite Cronwell (S.E. Head Researcher). Onboard, the pair had found 108 life-forms. The first of them (a pillar, shown as #00, later renumbered and known as #01) was an adult. Eight (later known as #02-09) were embryos. 99 others (later known as #10-108) were fertilized eggs. The adult and the eight embryos are also known as the "Single Numbers". Unwinged Single Numbers are equal or even more powerful than their winged counterparts. Scarletti has released the Sekirei out into Oasis as part of a game, pitting sekirei against one another. The roleplay begins in stage one where only a few of the sekirei have been winged. They have been released into the world in order to find a suitable ashikabi. The Sekirei becomes even more powerful and is able to do a prayer. A prayer is a special attack (be it physical or magical) that can only be done once winged. Sekirei under the same Ashikabi can also merge their techniques. Sekirei have a wing like tattoo on their upper back.

Ashikabi are humans with unique genes that enable them to empower the Sekirei destined to serve them. Through an exchange of saliva between a Sekirei and its Ashikabi (usually done through a kiss), a Sekirei without wings can become a "winged" Sekirei. The power of an Ashikabi can be determined not only by the number of the Sekirei he or she has in his or her disposal, but by the bond between them. If an Ashikabi dies, all of the Sekirei winged by him or her will also perish. Sekirei can be the same gender as their ashikabi.
